The series revolves around four Lucknow-based female police officers who work at a Mahila Police Station which consists of S.H.O. Haseena Mallik, S.I. Karishma Singh, Head Constable Pushpa Singh, Constable Santosh Sharma, and Constable Cheeteshwar Chaturvedi. They are helped by a prisoner-turned-spy Billu Champat. Karishma Singh is short-tempered while Haseena Malik is emotional in solving cases which sometimes creates a conflict between them.

Soon Karishma's twin sister Kaushalya aka Kareena who in her bid to become an actress fled with money saved by their father for Karishma's admission in police force arrives back to Lucknow to play a female cop in her upcoming movie and in her guilt and attempt to patch up with her sister asks Karishma to train her with Police tactics while Haseena asks them to swap their places in an attempt to make them realize difficulties faced in each other's jobs.
